How to prepare NGM (Nematode Growth Medium)?
l 2g peptone, 16g agar and 2.4 g NaCl placed in a clean 1000ml glass flask,
l Add 20ml 1M phosphate buffer added, with distilled water mixed to 800ml (the mixture is the initial culture medium).
l High pressure steam sterilization carried out at 120℃ for 30min
l Then cooling in a 55℃ water bath.
The preparation method of 1M phosphate buffer solution:
l 108.3g KH2PO4 and 35.6g K2HPO4 dissolved in 1000ml distilled water, and adjust the pH value to 6.0.
l 0.8ml of 5mg/ml cholesterol (dissolved in ethanol and not sterilized) and 0.8ml of 1M MgSO4 and 1M CaCl2 sterilized under high pressure were added in sequence, and the complete culture medium was obtained by shaking well.
